pH-responsive polymeric micelles with acid-labile ketal cross-link for drug delivery |

Micelles of amphiphilic block copolymer, poly(polyethylene glycol methyl ether methacrylate)-block-poly(styrene-alt-maleic anhydride)-block-poly(styrene) (PEGMA-b-PSMA-b-PS), prepared via the reversible addition-fragmentation chain transfer (RAFT) polymerization were cross-linked by amidation process of a degradable cross-linker and carboxylic acid group functional block copolymer. High doxorubicin (DOX) loading capacities (70% wt) were obtained. Core cross-linked micelles illustrated good stability under physiological condition but disassociated immediately under acidic pH. TEM images showed spherical shapes of DOX loaded cross-linked micelles with average hydrodynamic diameter of 67 nm. The DOX release experiments indicated a rapid DOX release at pH 5.0 compared to pH 7.4. The MTT assay demonstrated that the DOX loaded cross-linked micelles exhibited a high antitumor activity towards HepG2 cancer cells. |
