Recently, A study on intranasal delivery of vaccine progressed actively because of lack of pain and ease of administration. We studied on preparation of nanoparticle delivery system using biodegradable polymer, poly(lactide-co-glycolide)(PLGA) and characteristics of them. Two kind of PLGA nanoparticles were prepared by spontaneous emulsification solvent diffusion (SESD) method using 2% sodium dodecyl sulfate (SDS) as a anionic surfactant and 2% polyethylene-co-polypropylene diblock copolymer(Lutrol F68) as a nonionic surfactant. The vaccine was labeled with NHS-fluorescein. The labeled vaccine was coated on the surface of nanoparticles by ionic complexion. Characterization of nanoparticles for vaccine coating were analyzed by ESCA, ATR-IR and the size and zeta potential of nanoparticles were determined by particle analyzer. Coating amount of vaccine on the surface of nanoparticles was measured by fluorescence spectroscopy. The surface of nanoparticles was charged with anion and the size was lager of anionic nanoparticle than of nonionic nanoparticle. Also, zeta potential was higher of anionic particle than of nonionic particle.
