Rapid Amplification of Nucleic Acids by Photothermal Heating and Concentration in Plasmonic Cavity |

Fast detection through rapid nucleic acid (NA) amplification are continuously required in the recent Pandemic situation. Conventional polymerase chain reaction (PCR) uses thermocycler and takes about an hour to complete reactions. Here, we suggest a plasmonic cavity (PC), a spherical polymeric structure integrated with gold nanoparticles, as a faster and simpler NA amplification platform via light-induced heat generation by photothermal conversion. First, we investigate that the temperature is maintained to form proper conditions for recombinase polymerase amplification (RPA) and the polymerization reaction is accelerated by thermal gradient-driven concentration of reactants using the PC. Second, we observed the high photothermal conversion efficiency and rapid thermal cycle operated by the pulsed light source reduces the NA amplification time during PCR. As a result, the target DNA can be rapidly amplified to detectable levels in the PC under the light illumination conditions. |
