Polydiacetylene (PDA) liposomes have unique properties that allow liposomes to change color from blue to red and emit fluorescence in response to external stimuli. In this study, we demonstrate an antibody-based biosensor using PDA liposomes for detection of allergy diagnosis by hIgE. The surface of immoblized PDA liposomes were functionalized with hIgE monoclonal antibodies and the fluorescent signals were detected after antigen-antibody immunoresponse. However, fluorescent signals were slightly increased depending on the target protein concentration when polyclonal antibody conjugated with HRP were used as an amplifying agent after primary immunoresponse. After secondary immunoresponse, HRP-biocatalyzed oxdiation of DAB in the presence of H 2 O 2 formed precipitation of an insoluble product. As a result, a PDA liposome biosensor could detect effectively the hIgE and the sensitivity was evidently increased higher than the primary response. |
