Paclitaxel is a unique tubulin-interacting agent. In spite of paclitaxel's promising clinical drug activities, the drug has presented problems because of it's severe insolubility in water. Also, Paclitaxel has a special mechanism of action that inhibits SMC proliferation and migration. So, it has been applied to the surface treatment of stent for crown artery disease. But, restenosis and thrombosis are still problems in angioplasty. We have developed a series of water-soluble paclitaxel prodrug, based on PEG conjugates with a newly developed self-immolating group which gave a rapid hydrolysis rate to convert a prodrug into a parent drug without any reduction in drug efficacy. And, we have investigated its properties. In this study, we intend to induce an excellent anti-thrombosis action and a new drug delivery system based on a harmonized hydrophobicity/hydrophilicity ratio by developing a drug-eluting stent having a water-soluble paclitaxel prodrug. |
