The purpose of this study was to characterize and evaluate bone morphogenetic protein 2(rhBMP 2) sustained releasing ionic nano complex in vitro. The physico-chemical properties of ionic nano complex were measured. The protein conformational integrity of complex was investigated with FT-IR, CD and in vitro release study. The bioactivity of rhBMP2 within ionic nano complex was evaluated using C2C12 myoblasts. The particle was spherical with smooth surface and mean particle size, PI of the complex were 326 ± 149 nm and 0.2~0.3, respectively. The high resolution optical microscope, FT-IR and CD revealed that sustained releasing ionic nano complex system was successfully fabricated. The present system released rhBMP2 for 7 days and the bioactivity retained capable of differentiation to osteoblasts from undifferentiated C2C12 myoblasts. The present ionic nano complex system has shown potential possibilities for the sustained release of BMP2 and bone regeneration. |
