Expression of multimeric proteins from a single vector is a highly attractive method in broad fields of biomedical applications. In this study, we have developed in vitro transcribed (IVT) mRNA, incorporated with 2A peptide sequences, to express three fluorescent proteins (GFP, RFP and BFP). The 2A peptide sequences incorporated within IVT mRNA have obtained interest with their short length and high cleavage efficiency that induce the multiple protein expression. The IVT mRNA can be translated in the cytoplasm without entering into the nucleus, while a conventional DNA vector require to be transcribed after delivering into the nucleus. Thus, the efficiency of protein expression can be increased by the rapid translation of mRNA transcript in cytoplasm. We expect the developed multicistronic IVT mRNA system can be further utilized in various field of pharmaceutical sciences including mRNA vaccine research. |
