제목 A Thermosensitive and Biodegradable Chitosan/Oxidized Cellulose Nanofiber (TOCNF) Hydrogel System with Antifibrinolytic Agent Against Peritoneal Tissue Adhesions.
초록 Peritoneal tissue adhesion formation is a frequent problem after major abdominal surgery. To prevent peritoneal adhesion there have been various attempts by using mechanical barriers. In this study, an effective injectable thermosensitive hydrogel system was developed by combining various concentration of chitosan (1%, 2%, 3% & 4% w/v) and tempo-oxidized cellulose nanofiber (TOCNF) with antifibrinolytic agent mitomycin C for anti-adhesion application. These hydrogels can undergo sol-gel (4℃-37℃) transition at body temperature (37℃) through interactions between chitosan and β-glycerophosphate. TOCN ensured mechanical stability of the hydrogel and increased porosity. Higher amounts of chitosan resulted in faster gelation time with TOCNF. 4% chitosan with 1% TOCNF was free-flowing, capable of faster gelation time and injectable at ambient temperature. Chitosan has hemostatic properties and mitomycin C (MMC) prevents fibrinolysis which ensure antiadhesion after peritoneal surgery. These hydrogels with TOCNF showed improved biocompatibility both in vitro and in vivo rat model compared to only chitosan hydrogel. Both RBMSC cells and L929 fibroblast cells showed biocompatibility towards these hydrogel system with mitomycin C in vitro. Thus, TOCNF containing 4% chitosan with MMC bio compatible hydrogel system could be a potential candidate for adhesion barrier.
