제목 Effects of immobilized heparin and L-arginine on pericardial acellular matrices
초록 Biomaterials play a critical role in the engineering of new functional tissues for the replacement of lost or malfunctioning tissues. The drawback of matrices include durability, immunogenicity and calcification. In this study, to overcome these problems, acellular matrix of bovine pericardium(ABP) was chemical modified by the direct coupling of L-arginine(Arg) or heparin(Hep) containing amino groups, after glutaraldehyde(GA) cross-linking. The effects of Arg or Hep coupling on durability and calcification of ABP were investigated. Arg has been known to have interesting bioactivity such as antiplatelets activity, angiogenesis, wound healing promotion, nerve regeneration, cell growth and interactions of circulatory blood cells with the vessel wall. It is well known that Hep coupling onto the polymer surface have improved blood compatibility of blood contacting surfaces. Covalently bou,nd biomolecules are expected to act as space filler and help decrease calcification and increase antithrombogenicity. In addition, this modified matrix can be a good candidate for tissue engineering scaffold due to the bioactivity of Arg or Hep. The thermal and mechanical properties of Arg or Hep treated ABP were investigated by measuring shrinkage temperature and tensile strength. Digestion resistance to collagen enzyme, calcification behavior, and cell viability of modified matrices were evaluated in vitro and in vivo.
