Preparation of Biodegradable Elastic Hydrogels Based on Glycol Chitosan and PLGA-PEG-PLGA Triblock Copolymers for Tissue Expander Application |

In the previous work, various biodegradable PLGA-PEG-PLGA/PEG hydrogels having lag time before swelling were synthesized for tissue expander application. Although the delayed swelling was demonstrated, the hydrogels were mechanically weak and disintegrated into small particles during degradation. In this study, new hydrogels were synthesized via radical crosslinking reaction of vinylated glycol chitosan(GC) and PLGA-PEG-PLGA diacrylate. Various PLGA-PEG-PLGA triblock copolymers were synthesized and modified to have terminal acrylate groups. The vinylated GCs were synthesized by coupling reaction of glycidyl methacrylate(GMA) with GC. The number of vinyl group conjugated to GC could be controlled by varying the feed ratio of GMA to GC. In vitro degradation tests showed that their swelling and degradation properties could be controlled by varying the chemical composition and their physical stability was preserved even after degradation due to the presence of non-degradable GC networks. |
