Development of Chitosan/ β-Tri calcium phosphate microsphere loaded hydrogel as bone tissue engineered therapeutics |

For irregularly shaped skeletal defects, injectable scaffolds are more desirable for bone repair and regeneration. The aim of this study is to develop a biodegradable microsphere based thermo-sensitive hydrogel for bone regeneration. Different types of cross-linked microspheres, consisting of Chitosan and β-tricalcium phosphate (β-TCP) with different diameters are fabricated via water-in-oil (W/O) emulsion method. The effective thermo sensitive hydrogel is fabricated comprising methylcellulose (MC). Scanning electron microscopy (SEM) images suggested microsphere diameter is within a range of 100-400 micron. Also, the smoothness of microsphere is inversely proportional to chitosan while microsphere diameter is directly proportional to β-TCP concentration. Spectroscopic and crystallographic analyses reveal perfect assimilation of Chitosan with β-TCP and MC gel. The gelation time of hydrogel increases upon decreasing MC. In addition, higher β-TCP and MC, lower degradation of hydrogel is obtained. Water-soluble tetrazolium (WST) salt assay shows that the hydrogels (2% and 3% MC) are nontoxic while higher chitosan containing microsphere (3%) are toxic to MC3T3E1 pre-osteoblastic cells. The microsphere loaded samples were compatible enough to differentiate rat bone marrow mesenchymal stem cells and express the preliminary osteoblast marker (ALP) after 14 days. This study demonstrates that this safe and potential injectable system can be applied finally to prevent osteoporosis as target selective agent. |
