제목 | Bioinspired Surface-Engineering of Stem Cell Spheroids with Dexamethasone-Eluting Depots Promotes Bone Regeneration in Murine Calvarial Defect Model |
초록 | Three-dimensional cell culture and bioactive molecule co-delivery with cells have been proposed to enhance cell potency. Here, we surface-engineered mesenchymal stem cell (MSC) spheroids with cell-attachable polydopamine-coated microspheres that release dexamethasone to modulate the fate of MSCs on osteogenesis. Macroporous fibrin scaffold was used to deliver these spheroids. The conjugation was rapid and cell-friendly. DEXA depots significantly enhanced osteogenic differentiation of spheroids. The conjugated particles were stably anchored on MSCs during cell migration. Importantly, DEXA depots facilitated the bone regeneration after implanting the conjugated spheroids into a calvarial defect of C57BL/6 mice. Surface-engineered MSC spheroids with drug depots created a potential for achieving better performance of MSCs. |
