Tumor Imaging by NIRF and PET with Using NanocarrierMade of Polypeptides and Polydepsipeptides |

Micellar or vesicular molecular assemblies with diameters from 10 nm to 200 nm are expected to accumulate in cancer sites due to the enhanced permeation and retention (EPR) effect. Sarcosine is biodegradable amino acid, which is metabolized by endogenous sarcosine hydrogenase. Amphiphilic block polypeptides and polydepsipeptides, which are composed of a hydrophilic block of poly(sarcosine) and a hydrophobic helical block of poly(glutamate) or poly(lactide), were synthesized. These amphiphilic block polymers formed micelles and vesicles according to the balance of the two blocks. The micelle was labeled with ICG to show selective imaging of tumor especially with high tumor/liver ratio. Lactosome labeled with 18F was also successful in tumor imaging with high tumor/muscle ratio. These nanocarriers are useful for tumor imaging due to the EPR effect. |
