Hydrophobically modified chitosan for application in photodynamic therapy of cholangiocarcinoma |

Chitosan is regarded as a biocompatible polysaccharide. It has been reported to enhance delivery of bioactive agents across the nasal or mucosal layer. The cationic properties of chitosan offer chances to conjugate functional groups and incorporate anionic drugs for targeted drug delivery. We prepared hydrophobically modified chitosan using ursodeoxycholic acid (ChitoUDCA) for photosensitizer (PS) delivery to tumor cells. PS-incorporated ChitoUDCA nanoparticles were prepared by simple mixing of anionic PS and ChitoUDCA and then dialysis. Nanoparticles have less than 300 nm particle size and spherical shapes. When PS or PS-incorporated ChitoUDCA nanoparticles were treated to cholangiocarcinoma cells, ChitoUDCA enhances delivery of PS to tumor cells and then enhanced ROS level rather than PS itself. ChitoUDCA showed higher phototoxicity against tumor cells than PS itself. We suggest that ChitoUDCA is a promising vehicles for PS delivery. |
