제목 Poly(Amino Acid) based adjuvants containing squalene for enhancing both cell-mediated and humoral immune responses against influenza virus
초록 Adjuvants improve vaccine efficacy by accelerating and prolonging immune response and also by lowering the required dose and reducing side-effects. Currently, there is major research effort to develop powerful adjuvants for purified or synthetic recombinant vaccines that are safer but less immunogenic. Particularly, adjuvants that enhance cell-mediated (Th1) immunity are in great demand but very rare. In this study, we have developed a novel squalene-based cationic poly(amino acid) adjuvant (CASq) which elicits both cellular (Th1) and humoral (Th2) immune responses. CASq was demonstrated to promote cellular uptake of viral antigen and stimulate macrophages, leading to active production of IL-12. Further, co-administration of inactivated pdm H1N1 vaccine with CASq significantly increased generation of antigen-specific antibodies and T cell immune responses in mice, and resulted in complete prevention of disease symptoms and protection against lethal infection.
