제목 | 3D culture platform formed with lung decellularized extracellular matrix for small cell lung cancer cells |
초록 | Small cell lung cancer (SCLC) is highly invasive and resistant to most anti-cancer drugs, resulting in low survival rates. To develop a novel therapy, a suitable in vitro model to investigate physiology SCLC is essential as current drugs are limited to chemotherapy. To form a 3D culture matrix, porcine lung extracellular matrix (ECM) was decellularized and modified with methacrylic moieties for chemical crosslinks. Then, to compensate for the loss of hyaluronic acid (HA) in native tissue, HA was added to the ECM-based hydrogel. In the matrix, SCLC cells (NCI-H69) proliferated and formed clusters, and the presence of HA increased the drug resistance. Moreover, matrix stiffness affected cell phenotypes. Our findings reveal that the lung ECM-based 3D culture is a promising model to elucidate SCLC physiology.
