제목 In Vitro Evaluation of Nerve Regeneration of Muscle-Derived Stem Cellsby NGF-Loaded Bioactive Bulking Agent
초록 Urinary incontinence, which is defined as the involuntary loss of urine, may be caused by uretheal hypermobility and/or intrinsic sphincter deficiency. Injectable bulking agents have been used in the treatment of urinary incontinence. However, injection volume decrement caused by particle migration and resorption in the body has been a main problem. In this study, we prepared growth factor (NGF)-immobilized porous polycaprolactone (PCL)/Pluronic F127 beads as an injectable and bioactive bulking agent which can provide bulking effect and lead to regeneration of the defect nerves, and thus stimulate smooth muscle around urethra for the effective treatment of urinary incontinence. The morphologies and growth factor release behaviors of the beads were investigated by SEM and ELISA kit, respectively. The neuronal differentiation potential by the growth factor-loaded porous beads was also evaluated using muscle-derived stem cell as a model cell (Grant sponsor: NRF-2011-0001804).
