Rational design of multi-functional non-viral gene delivery to specific tissues |

The delivery of therapeutic genes to neurons has the potential to treat neurological disease and spinal cord injury. Nonviral vectors are an attractive alternative in that they offer improved safety profiles compared to viruses and can be targeted to specific neuronal subpopulations. However, most nonviral vectors suffer from significantly lower transfection efficiencies than neurotropic viruses. The development of minimally-invasive, non-viral gene delivery vehicles for the central nervous system (CNS) is an important technology goal in the advancement of molecular therapies for neurological diseases. One approach is to deliver materials peripherally that are recognized and retrogradely transported by motor neurons toward the CNS. This approach, especially tissue-specific targeting technique, can also be applied for gene therapy of cardiovascular disorders. |
