Polymers that have properties such as biocompatibility and stimuli-responsive degradability are highly desirable as smart drug delivery carriers. Herein we report a novel redox degradable polymer based on hyperbranched polyglycerol. First of all, we designed a new monomer, 2-{2-[2-(oxiran-2-ylmethoxy)ethyl]disulfanyl}ethanol (SSG), and synthesized PSSG bearing disulfide bond within a backbone of the monomer structure. The disulfide bond is a redox-responsive linkage and could be cleaved upon treatment with reducing agent, leading to selective degradation of polymers in cancer cells. A series of homo- and copolymers were carefully synthesized with controlled molecular weights and molecular weight distributions. Degradation property of the polymers was studied with GPC by observing molecular weight change before and after treatment with reducing agent like DTT. In vitro cytotoxicity test also demonstrated that these new polymers possesses an excellent biocompatibility. |
