The aim of this study was to develop a culture medium to minimize the formation of ferric ion precipitates, jarosite, and maximize ferrous ion oxidation rate by the chemolithoautotrophic bacteria, Acidithiobacillus ferrooxidans. For this purpose, statistical medium optimization was performed using a central composite design of the response surface methodology (RSM). Two responses of ferric ion concentration and ferrous ion oxidation rate were predicted with a cubic model of four factors, the initial concentrations of KCl, MgSO4·7H2O, and (NH4)2HPO4 and the initial pH of the medium. As a results of RSM, it was predicted that the ferric ion concentration and iron oxidation rate would be maximized when the initial pH and initial concentrations of KCl, MgSO4·7H2O and (NH4)2HPO4 were 1.791, 0.134g/L, 0.161g/L, and 1.547g/L, respectively. The prediction was validated by batch culture of A. ferrooxidans under optimized conditions and showed a 95% and 130% of consistency on each response. |
