Among the techniques used to detect for multiple miRNAs, colorimetric detection of miRNA using encoded hydrogel microparticles is considered as one of the promising technique for on-site practical application owing to their large multiplex capacity and inexpensive and simple analysis. However, it is still limited due to the assay time, particularly the colorimetric reaction time, is too long, which makes it difficult to be practically applied and undermines the detection accuracy. In this work, we present a rapid colorimetric assay based on encoded hydrogel microparticles with a significant decrease in the colorimetric reaction time by two factors: 1) increase in the number of enzymes bound to hydrogel microparticles via post-synthesis functionalization method, and 2) elevating the enzyme reaction temperature during colorimetric labelling. We validated comparable sensitivity for three miRNA targets and multiplex miRNA detection assay. |
